Ziele, Inhalte und Methode der Lehrveranstaltung

The aim of this practicum is to produce and purify recombinant proteins for characterization by NMR spectroscopy.
Through the course of this practicum, the students will gain an overview and hands on experience of the methods of recombinant protein expression. They will use state of the art techniques that are used in academia and industry with applications in biochemistry, structural biology, protein engineering, biotechnologies, pharmacy etc.

Art der Leistungskontrolle und erlaubte Hilfsmittel

The students will be assessed on their active participation during the course of the practicum as well as through a final report.

Mindestanforderungen und Beurteilungsmaßstab

In order to attend the course, the students needs to have basic laboratory experience as well as a superficial knowledge of protein biochemistry.

Escherichia coli is one of the organisms of choice for the production of recombinant proteins. Its use as a cell factory is well-established and it has become the most popular expression platform. For this reason, there are many molecular tools and protocols at hand for the high-level production of heterologous proteins, such as a vast catalog of expression plasmids, a great number of engineered strains and many cultivation strategies. We review the different approaches for the synthesis of recombinant proteins in E. coli and discuss recent progress in this ever-growing field.
